Pertinent twitter quote from Ron Gilbert about his approach to the new game: "MI3 doesn't fall out of canon. We were very careful about that. Murray is in this game." So it sounds like his approach has softened somewhat here, which I personally welcome. I'm interested in what Ron does coming back to MI, but I don't want that to be totally at the expense of the other stuff -- which was of variable (but sometimes very good) quality. So I'm glad that this announcement comes also with a bunch of announcements about other people involved like Dave Grossman, the 3 ex lucasarts composers and some other pretty canny choices like Khris Brown on casting and voice direction. As far as I can figure out the game is a follow up to Monkey Island 2, but somehow without completely destroying the canon of everything that followed. Not sure how that works, since Murray is met (and created) in the early parts of MI3, but perhaps we're into some sort of time/dimension shenanigans. That wouldn't be entirely inconsistent with MI lore.

Murray the demonic talking skull is in the trailer for Return to Monkey Island and he wasn't introduced until Curse of Monkey Island. Other great Ron Gilbert franchises include Deathspank and Putt Putt.
