If Princess Cadence had chosen the ove route, then that must mean the ove route had failed (assuming she did try). So perhaps the friendship route may be the better choice? I suggest going to see Rarity. Applejack is the element of honesty. But her honesty can be blinded by bias. So perhaps she's not the best choice in that regard. I suggest that the first thing you ask is if Cadence did in fact try to deal with the situation to confirm that assumption. As for why no males have approached Twilight may be because they are intimidated by her princess status.

Talk to Rarity. Apple Jack would be an unwise decision mostly because of the implications so far. To say Twi may be right could severely damage her & Spikes relationship, true or not. Speak to Rarity first, then, if Apple Jack is still to be considered, pursue a second conversation with Spike first as he would understand the accusation. (Just my argument against Apple Jack at this time, a thought of warning.)

Everyone is saying Rarity, but I maintain that AJ would be the better option. In the first place, being directly related to the issue at hand, she would have a better view of how Twilight's yearning has affected Spike, give us a more accurate timeline, and I remember there being hints that she was not opposed to the idea of polygamy, so long as it was discussed with her first. Rarity can gossip, but what she has seen may not be true, or even relevant.
