It has a psychological effect on humans, people cant comprehend that someone like Superman would be walking amongst them, so when they see Clark, with the glasses, they see the most normal guy in the world. in this scenario, you would NEVER guess that Clark is Superman, that thought would never cross your mind, cause looking at him, you would be seeing the most normal average guy, and not the most powerful being in the world. so Clark telling Lois to really look at him, Lois is crossreferencing Superman and Clark in her mind, allowing her to finally see who he is
